Affinity maturation AIMProve™
Following clonal isolation of targeted B cells using AIMSelect™, the antibodies produced by these cells can be further matured to improve their affinity for the target using the time-efficient, exvivo AIMProve™ process. AIMM’s cloned B cell lines express the enzyme activation-induced cytidine deaminase (AID). AID generates mutations in a B cell’s immunoglobulin genes. By selecting B cells expressing antibodies with increased affinity for the target, it is possible to progressively enhance the affinity of antibodies produced by the B cell. Utilizing labeled antigen, AIMProve™ can rapidly generate sub-clones of antigen-specific B cells that produce antibodies with exceptional affinities for an antigen. The same process can be applied to improve the stability and manufacturability of a promising candidate.
